Principles and Practices of Phase Plate Cryo-Microscopy 

Cryo-electron microscopy (cryo-EM) is undergoing a technological revolution that has moved the field from sub-nanometer resolution ‘blobs’ to atomic resolution structures. However, there remain challenges associated with imaging low contrast, radiation sensitive cryo-preserved specimens. ‘Enhanced’ phase contrast (PC) transmission electron microscopy (TEM) methods were developed to overcome these complications. I will present the theory behind the technologies and results showing the success of using phase plate methods for cryo-EM investigations of biological complexes.
